Electrophoresis and DNA Fragment Length Determination Biotechnology Education Kit

Restriction enzymes cut DNA at specific base sequences, and their discovery in the late 20th century rapidly expanded the field of molecular biology. In this activity, students run agarose electrophoresis on three lambda DNA samples pre-cut with HindIII, EcoRI, or both, and see how each enzyme affects the DNA differently.

Students learn to set up an agarose gel, load DNA samples, stain the gel to visualize the bands, and determine the sizes of unknown DNA fragments. The DNA samples need no refrigeration or freezing, and the prepared agarose simply melts in a hot water bath or microwave. TBE buffer and DNA stain are included.

• 200 ml Prepared Agarose, 0.8% Solution
• 500 ml Tris Borate ETA Buffer, 5X
• 60 ml DNA Stain, 20X
• 100 μl Lambda DNA/EcoRI Digest with Loading Dye
• 100 μl Lambda DNA/HindIII Digest with Loading Dye
• 100 μl Lambda DNA/EcoRI,HindIII Digest with Loading Dye
